Anti-human leukemia hairy cell
  

All IHC stains will include a positive control tissue

  
  • Most useful in distinguishing Hairy Cell Leukemia (HCL) from other low grade B cell lymphomas. DBA.44 reacts consistently with tissue infiltrates of HCL (97%) with positivity present in 50-75% of cells. 10% of other low grade B lymphomas may be reactive. About 35% of high grade B cell lymphomas show strong positive staining (i.e. centroblastic and immunoblastic types). Occasionally, some pleomorphic T cell lymphomas show reactivity in scarce large cells
  • DBA.44 is a useful marker in a panel approach (DBA.44, CD20, MB2, and anti-TRAcP) for Hairy Cell Leukemia
  • Reactivity in normal tissues:
    • Reactive lymph nodes and spleen: reacts with membrane immunoblasts outside lymphoid follicles. A few germinal centers may be positive in some lymph nodes
    • Thymus: occasional medullary lymphocytes are stained
    • Subpopulation of macrophages (which have phagocytized the antigen) are found to be reactive (cytoplasmic)
    • Reacts with very few small B lymphocytes in normal and reactive bone marrow
    • Cross-reactivity with lung alveolar lining cells, kidney tubules, some endothelial cells and salivary duct cells

  • Reacts with a subset of B lymphocytes localized in the follicular mantle zone

 
Staining patterns

  • Cell surface membrane (may be cytoplasmic)
  • Some immunoblasts outside lymphoid follicles in reactive lymph nodes and spleen may show a dot-like paranuclear staining
