  • All IHC stains will include a positive control tissue
  
  • Strong expression for this marker is seen in the epithelial/glomeruloid structures and blastemal components of Wilms' tumors
  • WT1 is highly specific for DSRCT (desmoplastic small round cell tumors), and identifies the presence of t(11;22)(p13;q12)
  • WT1 is found in granulosa cell tumors of females, and in sertoli cell tumors of males
  • WT1 protein is usually present in mesothelial cells (and mesothelioma cell lines), and is not seen in lung cancer cell lines (some lung cancers have been reported as positive); 54/56 mesothelial specimens (normal and hyperplastic mesothelium) were positive for WT1
  • WT1 is usually present in epithelial mesotheliomas (72%), and less frequently in sarcomatoid mesotheliomas
  • ALL, AML and blast crisis of CML are often positive for WT1; chronic leukemias are WT1 negative (WT1 is not seen in lymphomas)
  • WT1 has also been identified in renal cell carcinoma, melanoma, and serous papillary ovarian cancer
  • Serous papillary carcinoma of the ovary is usually WT1 positive (93%), and staining can also be seen in inclusion cysts, and tubal epithelium (cervical and endometrial epithelium is WT1 negative)

Specifications

  • WT1 is a gene located on chromosome 11p 13 , and is involved in the induction of Wilms' tumor
  • The protein encoded by WT1 regulates transcription of other genes, and functions both as a transcriptional activator (with P53) and suppressor (it represses EGF-R and IGF-1R)
  • WT1 is expressed in normal mesenchymally derived tissues, included mesothelium, sertoli cells, decidual cells, granulosa cells, pleura, heart, foreskin, endometrium and glomerular epithelium
  • This antibody also can be expressed in myelocytic cells and in some leukemias
  • It is useful in the identification of mesothelioma, ovarian serous carcinomas, desmoplastic round cell tumors of childhood, certain acute leukemias and extramedullary myeloid tumors, and ovarian and testicular and stromal tumors

Staining pattern

  • Nuclear based staining (in Wilms' tumors and malignant mesothelioma)
  • Cytoplasmic staining (in some adenocarcinomas and some areas of desmoplastic stroma)
  • Perinuclear staining (seen in some tumor cells of malignant mesotheliomas)
