Determine abnormal T cell lymphoproliferative phenotype

Pathology orders only

EDTA whole blood

4 mL

1.0 mL

Immediatley following collection, mix sample thoroughly by gentle inverting 8 - 10 times, to prevent clotting

Lavender (EDTA), 4mL

Body fluid
Bone marrow
Cerebrospinal fluid (CSF)
Tissue

Varies; see Alternate collection instructions

Body fluid

  • Collect 20 mL (minimum 5 mL) of body fluid and place in a Falcon vial/tube - 15mL
    • Do not add anticoagulant or holding media
    • The volume required is dependent upon the white blood cell (WBC) count. Smaller volumes are sufficient if the cell count is high
  • Include representative slides (will be returned)

Bone marrow

  • Collect 3 mL (minimum 1 mL) in a sodium heparinized (Na hep) syringe
    • Specimens collected in EDTA are acceptable, but not preferred
  • Transfer to an anticoagulant free sterile tube
  • If unable to collect a bone marrow aspirate, a bone marrow core placed in RPMI with Hepes, Pen Strep and FBS is acceptable
  • Submit representative slides (will be returned)

Cerebrospinal fluid (CSF)

  • Collect 5 mL (minimum 1 mL) in a sterile vial/container.
    • Do NOT add any anticoagulant or holding media
    • The volume required is dependent upon the white blood cell (WBC) count. Smaller volumes are sufficient if the cell count is high
  • Send representative slides (will be returned)

Tissue

  • Immediately following collection, place the fine needle aspirate (FNA) or tissue in RPMI with Hepes/Pen strep/FBS -15 mL
  • Send representative slides (will be returned)

  • Improper label (unlabeled or mislabeled)
  • Wrong container (anticoagulant or solution)
    • Tissues submitted in saline or B+ fixative are not acceptable and will be rejected
  • Improper blood/anticoagulant ratio
  • Delay in transport
  • Improper storage
  • Inappropriate timing of collection
  • Interfering substances
  • Leaking container

Tubes and Markers:

Tubes:

T1, T2, T3*

T3 tube reflexed for an additional charge, only if immature or lymphoblastic features are detected in the T1 and/or T2 tubes

Markers:

CD2, CD3, CD4, CD5, CD7, CD8, CD10, CD16, CD19, CD45, CD56, CD57, HLA-DR, TCRαβ, TCRγδ, TRBC1

* If T3 tube is reflexed, additional markers include: CD1a, CD3(cytoplasmic), CD34, TdT(nuclear))
